Title: Large-scale CRISPR screening in primary human 3D gastric organoids enables comprehensive dissection of gene-drug interactions.
Screen Details:
- Screen Rationale : Increased/decreased sensitivity to cisplatin
- Cell Type : Gastric Tumor Organoid Model
- Cell Line : Organoid: TP53/APC knockout gastric tumor organoid line
- Phenotype : Response To Chemicals
- Condition : Cisplatin (1.6 µg/mL) [ChemSpider]
- Library : CRISPRa targeted library (Chen, 2025) | Type: CRISPRa | Format: Pool | Enzyme: Cas9 | Methodology: Activation
- Analysis Method : Mann-Whitney U test | Number of Hits: 10 | Full Dataset Size: 1,952
- Experimental Setup : Drug Exposure | Duration: 7 Doublings | MOI: = 0.3
- Significance : Authors used a complex method to determine significant results. Please refer to the source publication for more details. Significant results are indicated by '' in the HIT column of the table below.
Notes:
- A targeted CRISPRa screen was carried out in a human Tp53/APC double knockout gastric tumor organoid model to identify genes whose overexpression conferred sensitivity to cisplatin. The CRISPRa library targeted 1952 genes encoding DNA and RNA-binding proteins in the organoids. To call gene hits, a "screen score" was defined as | gamma z-score from negative control gene distribution | x -log10 p-value.
