Title: CRISPR activation and interference screens decode stimulation responses in primary human T cells.
Screen Details:
- Screen Rationale : Regulation of stimulation-responsive cytokine production
- Cell Type : T Cell [CL:0000084]
- Cell Line : CD4+ T-cells
- Phenotype : Protein/peptide Accumulation [APO:0000149]
- Condition : Interleukin-2 (IL-2) (500.0 IU/ml) [Uniprot]
- Library : Human CRISPR Activation Pooled Library (Calabrese) [AddGene] | Type: CRISPRa | Format: Pool | Enzyme: dCas9-VP64 | Methodology: Activation
- Analysis Method : MaGeCK | Number of Hits: 243 | Full Dataset Size: 18,930
- Experimental Setup : Cytokine exposure | Duration: 6 Days | MOI: ~ 0.5
- Significance : Authors have indicated that results with Score.2 (FDR) < 0.05 AND Score.5 (Log2FC) > 0.5 OR Score.4 (FDR) < 0.05 AND Score.5 (Log2FC) < -0.5 are to be considered significant. These results are indicated by '' in the HIT column of the table below.
Notes:
- Hit genes with a log2fc>0.5 and FDR<0.05 are enriched upon activation in a population sorted for IL-2-producing CD4+ T cells, suggesting the gene is a positive regulator of IL-2-production
- Hit genes with a log2fc<-0.5 and FDR<0.05 are depleted upon activation in a population sorted for IL-2-producing CD4+ T cells, suggesting the gene is a negative regulator of IL-2-production upon activation
- The screen duration (6 days) is calculated as the time from infection with the sgRNA library to harvest of the cells for FACS. Cells were not exposed to IL-2 stimulation for the full duration of the screen.
