Title: High-throughput screening of genetic and cellular drivers of syncytium formation induced by the spike protein of SARS-CoV-2.
Screen Details:
- Screen Rationale : Decreased syncytium formation
- Cell Type : Lung Adenocarcinoma Cell Line [BTO:0001911]
- Cell Line : A549-ACE2-Cas9-GFP1–10
- Phenotype : Syncytium Formation
- Condition : SARS-CoV-2 Spike
- Library : MinLibCas9 [AddGene] | Type: CRISPRn | Format: Pool | Enzyme: Cas9 | Methodology: Knockout
- Analysis Method : MaGeCK | Number of Hits: 9 | Full Dataset Size: 18,758
- Experimental Setup : SARS-CoV-2 Spike exposure (293T-spike-GFP11-P2A-mCherry cells) | Duration: 7 Days | MOI: = 0.3
- Significance : Authors have indicated that results with Score.1 (RRA rank) > 4.0 are to be considered significant. These results are indicated by '' in the HIT column of the table below.
Notes:
- A whole-genome CRISPR screen was performed to identify host-factors that enhance SARS-CoV-2 Spike-induced syncytium formation. Knockout of the top gene hits in A549-ACE2 receiver cells (A549-ACE2-SpCas9-GFP1-10 cells) inhibited syncytium formation when mixed with the WT spike-expressing sender cells (293T-spike-GFP11-P2A-mCherry cells).
