- Cell Type : Lung Adenocarcinoma Cell Line [BTO:0001911]
- Cell Line : Calu-3 [BTO:0002750, CELLOSAURUS:CVCL_0609]
- Phenotype : Protein Binding
- Condition : SARS-CoV-2 Spike-RBD (10.0 nM)
- Library : H1 CRISPRi (Horlbeck, 2016) | Type: CRISPRi | Format: Pool | Enzyme: dCas9-BFP-KRAB | Methodology: Inhibition
- Analysis Method : MAGeCK-iNC | Number of Hits: 252 | Full Dataset Size: 2,555
- Experimental Setup : SARS-CoV-2 Spike-RBD exposure
- Significance : Authors have indicated that results with Score.1 (MaGeCK Score) < 0.0 AND Score.2 (p-Value) < 0.05 OR Score.1 (MaGeCK Score) > 0.0 AND Score.2 (p-Value) < 0.05 are to be considered significant. These results are indicated by '' in the HIT column of the table below.
Notes:
- A focused CRISPRi screen targeting the druggable genome (2,300 genes) was performed to identify factors controlling SARS-CoV-2 Spike-RBD binding to human cells. Genes were knocked down by CRISPRi and those that decreased (negative score) or increased (positive score) Spike-RBD binding were identified, including ACE2 and regulators of ACE2.
