Title: A genome-wide CRISPR screen identifies host factors that regulate SARS-CoV-2 entry.
Screen Details:
- Screen Rationale : Increased resistance to virus
- Cell Type : Lung Cancer Cell Line [BTO:0000762, EFO:0002934]
- Cell Line : A549-ACE2
- Phenotype : Response To Virus
- Condition : Virus: SARS-CoV-2 Sdel (deletion in S1/S2 site of spike protein) (3.0 MOI)
- Library : Brunello (human) [AddGene] | Type: CRISPRn | Format: Pool | Enzyme: Cas9 | Methodology: Knockout
- Analysis Method : MaGeCK | Number of Hits: 34 | Full Dataset Size: 19,114
- Experimental Setup : Virus Exposure | Duration: 7 Days | MOI: ~ 0.3
- Significance : Authors have indicated that results with Score.1 (MAGeCK pos score) > 0.0 AND Score.2 (MAGeCK pos score) > 0.0 AND Score.3 (FDR) < 0.15 are to be considered significant. These results are indicated by ' ' in the HIT column of the table below.
Notes:
- A CRISPR screen was performed in A549 cells expressing the ACE2 receptor. The cell survival-based screen was carried out using the SARS-CoV-2 Sdel virus with a deletion in the S1/S2 site of the spike protein in order to identify genes that regulate endosomal entry or surface expression of the ACE2 receptor.
