Title: Genome-wide CRISPR screen identifies host dependency factors for influenza A virus infection.
Screen Details:
- Screen Rationale : Increased resistance to virus (host dependency factors)
- Cell Type : Non-Small Cell Lung Cancer Cell Line [BTO:0002553]
- Cell Line : A-549 [BTO:0000018, CELLOSAURUS:CVCL_0023]
- Phenotype : Response To Virus
- Condition : Virus: Influenza A (A/Puerto Rico/8/1934(H1N1) (5 MOI) [NCBI Taxid 211044]
- Library : CRISPRn targeted library (Hacohen, 2020) | Type: CRISPRn | Format: Pool | Enzyme: Cas9 | Methodology: Knockout
- Analysis Method : FDR | Number of Hits: 121 | Full Dataset Size: 1,000
- Experimental Setup : Virus Exposure | Duration: 16 Hours
- Significance : Authors have indicated that results with Score.1 (FDR) < 0.05 are to be considered significant. These results are indicated by '' in the HIT column of the table below.
Notes:
- Based on the results of the primary screen the authors performed a targeted secondary screen with 10 sgRNAs/gene for the top 1000 ranked genes from the initial screen
- Cells transduced with the sgRNA library and exposed to influenza were FACS sorted for expression of viral HA. Cells with low viral HA were sorted into an "uninfected" bin and sgRNAs enriched in this bin should represent host dependency factors.
