Title: Survival-based CRISPR genetic screens across a panel of permissive cell lines identify common and cell-specific SARS-CoV-2 host factors.
Screen Details:
- Screen Rationale : Increased resistance to virus
- Cell Type : Bladder Carcinoma [EFO:0000292]
- Cell Line : UM-UC-4 [CELLOSAURUS:CVCL_2749]
- Phenotype : Response To Virus
- Condition : Virus: SARS-CoV-2/SB3-TYAGNC (0.001 MOI)
- Library : Toronto Knockout version 3 (TKOv3) [AddGene] | Type: CRISPRn | Format: Pool | Enzyme: Cas9 | Methodology: Knockout
- Analysis Method : Quantitative genetic interaction (qGI) method (Chan, 2022) | Number of Hits: 1 | Full Dataset Size: 17,812
- Experimental Setup : Virus Exposure | Duration: 23 Days | MOI: ~ 0.3
- Significance : Authors have indicated that results with Score.1 (Differential score) > 0.0 AND Score.2 (FDR) < 0.1 are to be considered significant. These results are indicated by '' in the HIT column of the table below.
Notes:
- A genome-wide CRISPR screen was carried out to select for host factors required for SARS-CoV-2 cytotoxicity. Positive hits were identified using a modified quantitative genetic interaction (qGI) scoring method. None of the identified gene hits selected by SARS-CoV-2 exposure were identified as essential fitness genes in the accompanying cell-essential CRISPR screen in the same cell line. (Virus exposure for 23 days at MOI = 0.001.)
